Short-chain fatty acids (SCFA) are organic fatty acids with carbon atoms of 2-6, mainly including acetic acid, propionic acid, butyric acid, isobutyric acid, valeric acid, isovaleric acid, capric acid and isohexanoic acid. SCFA are mainly produced by fermenting indigestible sugars such as dietary fiber, resistant starch and oligosaccharides in the colon by beneficial bacteria such as lactobacilli and bifidobacteria.
Metabolomics is the study of metabolite changes in organisms, which can provide information on the metabolism of microorganisms in the gut. By detecting metabolites in various body fluids (e.g., urine, plasma, and feces), functional profiles of common metabolites of gut microbes and hosts can be obtained, which can help to study metabolic pathways affected by diseases or drugs associated with gut microbes and find relevant biomarkers as well as targets for drug therapy.
The gut microbiome can be altered by a variety of disease states, including metabolic and inflammatory diseases, pain syndromes, neurodegenerative diseases, and stroke. To date, only a few experimental studies have focused on the role of the gut microbiome in cerebral ischemia.
The Women’s Health in Neuroscience Program from the Department of Neuroscience and Experimental Therapeutics at the Texas A&M University Health Science Center investigated the association of reproductive aging with changes in gut microbial communities and metabolites at baseline and post-stroke in rats, and the effects of estrogen on stroke recovery in adult and reproductively aged female rats. In this study, Creative Proteomics provided SCFA analysis using the nanoLC-MS/MS platform. Stool samples collected from adult and middle-aged female rats at day 0 and 5 days after MCAo were analyzed for seven SCFAs, including acetate, butyrate, propionate, isobutyrate, valerate, isovalerate, and hexanoate. The results showed that reproductive aging in female rats induces dysbiosis of the gut microbiota that is exacerbated by stroke, leading to intestinal leakage and altered metabolites produced by the altered microbiota.
